RESUMEN

Spatial transcriptomics has revolutionized the study of gene expression within tissues, while preserving spatial context. However, annotating spatial spots' biological identity remains a challenge. To tackle this, we introduce Pianno, a Bayesian framework automating structural semantics annotation based on marker genes. Comprehensive evaluations underscore Pianno's remarkable prowess in precisely annotating a wide array of spatial semantics, ranging from diverse anatomical structures to intricate tumor microenvironments, as well as in estimating cell type distributions, across data generated from various spatial transcriptomics platforms. Furthermore, Pianno, in conjunction with clustering approaches, uncovers a region- and species-specific excitatory neuron subtype in the deep layer 3 of the human neocortex, shedding light on cellular evolution in the human neocortex. Overall, Pianno equips researchers with a robust and efficient tool for annotating diverse biological structures, offering new perspectives on spatial transcriptomics data.

Atmospheric aerosols have been found to influence the development of planetary boundary layer (PBL) and hence to aggravate haze pollution in megacities. PBL height (PBLH) determines the vertical extent to which the most pollutant effectively disperses and is a key argument in pollution study. In this study, we quantitatively evaluate aerosol radiation effect on PBL, as well as assessment of surface cooling effect and atmosphere heating effect. All the data are measured at a site of Beijing from 2014 to 2017, of which PBLH is retrieved from micro pulse lidar and aerosol optical depth (AOD) from sunphotometer. Case study shows qualitatively that relative high aerosol load reduces PBLH, and in turn causes a high surface PM2.5 concentration. We preliminarily reveal the influential mechanism of aerosol on PBL. The influence of aerosol on the radiation flux of PBL is analyzed, with the correlation coefficient (R) of 0.938 between AOD and radiative forcing of BOA (RFBOA) and R = 0.43 between RFBOA and PBLH. Also, AOD is found to negatively correlate with PBLH (R = -0.41). With the increase of AOD, the cooling effect of surface is enhanced, and further impede the development of PBL. Due to aerosol-induced reduction of PBLH, near surface PM2.5 concentration surges and presents an exponential growth following AOD. Then, it is speculated and testified that the relationship between SSA (single scatting albedo) and PBLH would be determined by the location of absorbing aerosol within PBL. The upper PBL absorbing aerosol may decrease PBLH, while the lower absorbing aerosol appear to enhance PBLH. The study probably can provide effective observational evidence for understanding the effect of aerosol on PBL and be a reference of air pollution mitigation in Beijing and its surrounding areas.

A neuroinflammatory response is commonly involved in the progression of many neurodegenerative diseases. Potassium 2-(1-hydroxypentyl)-benzoate (PHPB), a novel neuroprotective compound, has shown promising effects in the treatment of ischemic stroke and Alzheimer׳s disease (AD). In the present study, the anti-inflammatory effects of PHPB were investigated in the plasma and brain of C57BL/6 mice administered a single intraperitoneal (i.p.) injection of lipopolysaccharide (LPS). Levels of iNOS and the cytokines TNFα, IL-1ß and IL-10 were elevated in plasma, cerebral cortex and hippocampus after LPS injection and the number of microglia and astrocytes in cortex and hippocampus were increased. LPS also upregulated the expression of heme oxygenase-1 (HO-1) in the cortex and hippocampus. PHPB reduced the levels of iNOS and cytokines in the plasma and brain, decreased the number of microglia and astrocytes and further enhanced the upregulation of HO-1. In addition, PHPB inhibited the LPS-induced phosphorylation of ERK, P38 and JNK. These results suggest that PHPB is a potential candidate in the treatment of neurodegenerative diseases through inhibiting neuroinflammation.

Four new benzofuran-type stilbene glycosides and 14 known compounds including 8 benzofuran-type stilbenes and 6 flavonoids were isolated from the traditional Chinese medicine, Cortex Mori Radicis. The new compounds were identified as (9R)-moracin P 3'-O-α-l-arabinopyranoside (1), (9R)-moracin P 9-O-ß-d-glucopyranoside (2), (9R)-moracin P 3'-O-ß-d-glucopyranoside (3), and (9R)-moracin O 10-O-ß-d-glucopyranoside (4) based on the spectroscopic interpretation and chemical analysis. Three benzofuran-type stilbenes, moracin O (5), R (7), and P (8) showed significant neuroprotective activity against glutamate-induced cell death in SK-N-SH cells. In addition, moracin O (5) and P (8) also demonstrated a remarkable inhibition of the acetic acid-induced pain. The molecular docking with metabotropic glutamate receptor 1 (mGluR1) results indicated that these neuroprotective benzofuran-type stilbenes might be the active analgesic components of the genus Morus, and acted by mediating the mGluR1 pathway.

Twenty polycyclic polyprenylated acylphloroglucinols (PPAPs), including the new compounds hyperscabrones A-I (1-9), were isolated from the air-dried aerial parts of Hypericum scabrum. These compounds comprise seven different structural types. All structures were determined by NMR spectroscopic methods and both experimental and calculated electronic circular dichroism (ECD) spectra. The evaluation of their neuroprotective effects on glutamate-induced toxicity in SK-N-SH cells showed that compounds 4-7 exhibited significant neuroprotection at 10 µM. Additionally, compounds 3, 4, 7, and 9 showed moderate hepatoprotective activities against paracetamol-induced HepG2 cell damage at 10 µM.

RESUMEN

Alzheimer's disease(AD) is a chronic progressive neurodegenerative disease. The pathogenesis of AD is unclear, and it is presently incurable. Medicines currently available for AD treatment are only for improving the cognitive symptoms, but not able to stop or delay disease progression. Here, we summarized the interventions in early phases of AD in clinical trial. As a complex disease, AD is difficult to be restored through a treatment on a single target. Multi-target and cocktail drugs might be a strategy for development of AD therapies. In addition, AD is characterized by progressive neuronal loss in the cortex and hippocampus. The induction of neurogenesis by small molecule compounds has drawn attention in the AD field. The study of natural products in China is leading the way in the AD world. Numerous natural products have been identified for pharmacological effects on multi-targets in the regulation of neurogenic activity, which may open up a new avenue for AD treatments.
